-Key points of presentation-
We successfully purified photosystem I monomer and trimer from Acaryochloris, which has low energy chlorophyll d.
We suggested that the composition and orientation of low-energy chlorophyll d are different between photosystem I monomers and trimers.
The diversity of low-energy chlorophyll d may be important in understanding the evolution of Acaryochloris species.

◆Overview
A research group led by Associate Professor Ryo Nagao of Shizuoka University’s Faculty of Agriculture, together with Professor Kenren Shen of Okayama University and Unit Leader Nao Domae of the RIKEN Center for Environmental and Resource Science, has investigated the cyanobacterium Acaryochloris sp. NBRC 102871 (hereinafter referred to as We purified photosystem I (PSI) monomer and trimer from NBRC102871) and clarified their molecular properties.
Low-energy chlorophyll d was observed in both the PSI monomer and trimer by UV-visible spectroscopy and fluorescence spectra. However, their spectral properties were significantly different.
As a result of structural changes occurring around chlorophyll d due to the interaction between PSI monomers, it was suggested that different low-energy chlorophyll d was formed in the PSI trimer and monomer. This is characteristic of Acaryochloris species, which have diverse low-energy chlorophyll d.
Chlorophyll d is only found in Acaryochloris species. Other oxygenic photosynthetic organisms such as cyanobacteria, algae, and land plants do not have chlorophyll d.
We analyzed the PSI of Acaryochloris marina MBIC 11017 (hereinafter MBIC11017) in 2023, but we did not observe as low-energy chlorophyll d as NBRC102871.In this way, the PSI of Acaryochloris species, which is a special group of organisms, We found that the low-energy chlorophyll d that binds is diverse.
The research results obtained in this study will be important knowledge in understanding the evolution of Acaryochloris species. The results of this research were published in the international magazine “Photosynthesis Research” published by Springer Nature on June 27, 2024.

◆Researcher’s comment: Ryo Nagao, Associate Professor, Faculty of Agriculture, Shizuoka University In my laboratory, we study the differences in the apparent colors of photosynthetic organisms at the molecular level. The Acaryochloris species, which have chlorophyll d, are a truly mysterious group of organisms. Why didn’t they use chlorophyll a, which is used by other photosynthetic organisms? We conducted this research to clarify such a simple question. I think we have clarified part of this with our results.
